This thesis consists of the design and implementation of a sequential control for controlling the focus knob of a NIKON SMZ1000 stereoscopic microscope. Image stacking consists of taking photographs at different focal lengths, and then stacking the images to obtain a fully focused image. To achieve this, the focus screw moves the stereoscope head several steps sequentially and precisely. This process was programmed to work within a defined displacement and magnification configured by the user. Using a stepper motor, the knob will advance one step at a time until it reaches the final position. Between each step, a DSLR camera located in the upper trinocular will take the photograph. After completing the series, the final set of images can be stacked with various editing software such as Helicon Focus, Adobe Photoshop, or CombineZP. The sequential command is implemented using an Arduino Mega 2560 as the controller, a 2.1A NEMA 17 motor for movement, and a DVR8825 driver for motor control. The work was carried out in collaboration with Nikon Peru Microscopes, which provided technical expertise and formulas for selecting the suggested magnification step. The experimental results demonstrate user time savings of up to 65 %, improved image quality, and the elimination of human error caused by head movement or margin misalignment.
